MALDI-MSI Instruments

The NCMSI platform is one of the largest and most modern MALDI-MSI laboratories in Europe. It has two MALDI TOF-TOF mass spectrometers (Ultraflextreme, rapifleX, Bruker Daltonics; installed in 2014 and 2017), a dual source (MALDI/ESI) FTICR mass spectrometer (Bruker Daltonics, installed 2017), and one MALDI-q-TOF instruments – a Synapt G2-S HDMS with ion mobility separation (Waters Corp., installed in 2015).  The laboratory also has several MALDI matrix application instruments including two ImagePrep instruments (Bruker Daltonics; uses vibrational vaporization technology), one CHIP-1000 Chemical Printer (Shimadzu; uses piezoelectric spotting technology), and one TM-Sprayer Tissue MALDI Sample Prep System (HTX Technologies; uses heated spray technology).

Mass Spectrometers
MALDI-TOF-TOF, Bruker Daltonics
The ultrafleXtreme can perform 2 kHz speed in MS mode and 1 kHz in MS/MS mode with ultrahigh performance and flexibility for a broad variety of MALDI-MSI applications. Can analyze samples at near cellular spatial resolution (10-20µm). Requires tissue sections to be placed on conductive glass slides. Instrument installed and 2014.

rapifleX MALDI Tissuetyper – High speed and high spatial resolution. Up to 40 true pixels/s. smartbeam™ 3D laser for better pixel-to-pixel reproducibility that fires at a repetition rate of up to 10 kHz with a laser diameter of 5um. Requires tissue sections to be placed on conductive glass slides. Installed spring 2017.

ESI/MALDI FTICR MS, Bruker Daltonics

Dual source (MALDI and ESI) FTICR mass spectrometer (7T solariX 2xR). Operating with a kilohertz Smartbeam II laser. Resolving power of 450,000 at m/z 400. Installed spring 2017.

MALDI/ESI Q-Tof Synapt G2si, Waters Corp.
The MALDI SYNAPT G2-Si is powered by a solid-state laser with a repetition rate of 2.5 kHz to maximize the speed of the analysis. In addition, SYNAPT is well suited for analysis of tissue sections placed on regular microscope glass slides (non-conductive). Spatial resolution of 50-10µm. Instrument installed 2015.

Matrix Application and Tissue Cutting Instruments
TM-Sprayer, HTX-Technologies
The TM-Sprayer provides heat to the matrix solution to accelerate adsorption into the tissue, as well as a controlled flow of dry air or nitrogen to focus the spray and control drying time.

Using vibrational vaporization technology, ImagePrep provides reproducible matrix preparations for MALDI-MSI with a fully automated process.

This instrument delivers picoliter volumes of reagent to a tissue section surface using piezoelectric technology. To support the visualization of the droplet and the area of interest, the system has a camera and an in built scanner.
